Abstract

 

Osthole is a natural coumarin that was first isolated from several medicinal plants, including C. monnieri and A. pubescens. It is reported to induce a nonspecific elevation of cAMP and cGMP levels through its ability to inhibit phosphodiesterases, which leads to a modulation of calcium channel signaling and vasorelaxant effects.1 Osthole exhibits immunomodulatory, antioxidative, and anti-inflammatory properties, by regulating the expression of TNF-α, NF-κB, TGF-β, cyclooxygenases, leukotrienes, nitric oxide, ERK, and JNK. It also demonstrates antifibrotic effects in both hepatic and myocardial tissues and has been suggested to be a natural activator of defective delta F508-cystic fibrosis transmembrane conductance regulator chloride channel gating.
 

Cnidium monnieri is one of the most widely used traditional herbal medicines and its fruits have been used to treat a variety of diseases in China, Vietnam, and Japan. 350 compounds have been isolated and identified from C. monnieri, including the main active constituent, coumarins. In vitro and in vivo studies suggest that osthole and other coumarin compounds possess wide range of pharmacological properties for the treatment of female genitals, male impotence, frigidity, skin-related diseases, and exhibit strong antipruritic, anti-allergic, antidermatophytic, antibacterial, antifungal, anti-osteoporotic effects.

The objectives of this study were to determine the therapeutic effect of osthole, an active constituent isolated from Cnidium monnieri, in hyperlipidemic fatty liver rats and investigate the possible mechanism of the osthole treatment. Osthole exerts therapeutic effects on fat milk-induced fatty liver in rats, by regulating mRNA expression of the target genes of CYP7A, DGAT, L-FABP and FATP4 via increasing the PPARalpha/gamma mRNA expression.

The normalization of plasma alanine aminotransferase (ALT) has been proved to be a strategy for preventing the development of hepatocellular carcinoma in hepatitis C virus (HCV)-infection. Glycyrrhizin, a plant medicine, normalizes plasma ALT and prevents HCC. However, glycyrrhizin is administered intravenously and thereby chemical which is effective on oral administration is required. Coumarin compounds are active components of herbs used for the treatment of various diseases. The ability of coumarin compounds to lower plasma ALT were examined using mice concanavalin A-induced hepatitis and mice anti-Fas antibody-induced hepatitis. We conclude that osthole will become a leading chemical for synthesizing a compound which prevents liver cancer on oral administration.
