Description:
Paroxetine is a phenyl acridine compound that has the strongest ability to inhibit 5-HT compared to other 5-HT reuptake inhibitors. It has an inhibitory limit of 1.1 nM for [3H]-5-HT, 25 nM for fluoxetine, 6.2 nM for fluoxetine, 7.4 nM for clomipramine, and 100 nM for imipramine. Paroxetine inhibits 5-HT absorption. The mechanism increases the amount of 5-HT available for bioavailability in the synaptic cleft of nerve cells, thereby increasing 5-HT energy transmission and exerting antidepressant effects. Paroxetine can cause a decrease in 5-HT in platelets, suggesting that changes in 5-HT in platelets may reflect the effects of paroxetine on synapses.

Mechanism of action:
Paroxetine hydrochloride exerts its pharmacological effects by inhibiting 5-HT reuptake of brain neurons, and is more selective than tricyclic antidepressants (TCAs), fluoxetine, and sertraline, and rarely acts through other neurotransmitters. Low affinity for histamine H1 receptor, adrenergic alpha or beta receptor, dopamine D2 receptor, paroxetine hydrochloride does not act on NE receptors in the brain, suggesting that the drug has little sedative effect on cognitive processes or The impairment of psychomotor function is also small, and the cholinergic receptor affinity and cardiovascular adverse reactions are less than TCAs, and short-term or long-term treatment has no specific changes in hematology, biochemistry and urinary system.
